Purpose / Objectives

Main Objective
•To identify and select PRSV-CP transgenic line (s) with high level of resistance to PRSV

Specific Objectives :
•To multiply in vitro transgenic (T1) plants and their progenies (T2 to T4) for virus resistance testing and molecular analyses

•To conduct virus resistance screening to assess and confirm the PRSV reaction of selected plants and/or progenies in BL2 greenhouse/screenhouse

•To generate further molecular data of selected highly resistant PRSV event/line(s)

•To establish generational stability of gene insert and PRSV resistance trait of selected events and lines

Biosafety measures

•All plants and viable materials within the experimental area shall be accounted for.

•The proponents shall terminate all infected plants (showing severe yellowing and leaf deformation) and their in vitro counterparts that are no longer needed in data generation by chopping and burying the potted materials inside the screenhouse after treatment with bleach or steam sterilization for smaller volumes of cultured propagules. However, transgenic plants and their in vitro counterparts that will show higher level of resistnace reactions compared with the control will be retained for further observation.

•All farm tools and equipment (e.g. bolo, wheelbarrow) that will be used will be washed with tap water and disinfected with bleach solution and air-dried before returning them to the tool room.

•Disposal procedures will be conducted under the supervision of the BPI-PQS and UPLB-IBC representatives.

•Best management practices to prevent unintended PRSV infection inside the BL2 facility will be observed. These include keeping the screens undamaged, appropriate pesticide treatment to control aphid vector and other insect pest and diseases, and restricting physical movements of personnel.

•The proponents shall coordinate with the PQS-BPI for other requirements that they may impose, as necessary, according to their legal mandate.

Conditions for Approval

•The proponent should also specify when the plants will be challenged with the virus.

•The proponents must specify when plants deemed susceptible will be terminated (i.e. 8 weeks post inoculation (wpi)).

•The Gantt chart must specify when the promising lines will be terminated (e.g. after harvest of a specific number of fruits).
