Experimental Facility/Site Phenotypic analysis will be performed at IRRI CL4 and transgenic screenhouse. Seeds will be exported to C4 rice consortium members’ laboratories.
Purpose / Objectives

• Identify rice mutants with activation insertions in vicinity of C4-related genes from rice mutant collections.
• Screen for biochemical changes and lower CO2 compensation point in rice mutants.
• Screen for altered leaf anatomical features, vein density, bundle sheath cells in activation mutants.
• Determine genetic control of C4-related traits in mutants through co-segregation analysis of insertions and C4 characters.

Biosafety measures

• Activation lines received from Taiwan and Korea will be grown in the CL4 and transgenic greenhouses (CS01 and CS02), where a security system prevents access of unauthorized personnel.

• The plant materials, pots and soil for disposal from CL4 will be placed in sealed plastic bags and autoclaved at high temperature and high pressure. Plant material from transgenic screenhouse (CS01, CS02) will be ploughed back at the site or put in the pit to decompose.

• Transgenic seeds will be kept in designated area for storage of transgenic seeds in KJ Lampe Building, IRRI.

• Movement of materials will be done in compliance of all relevant biosafety and phytosanitary requirements of the NCBP.
